張雜谷提取物的抗氧化研究及蔗糖合成酶基因家族的生物信息學(xué)分析摘要:

本研究對張雜谷提取物的抗氧化能力及其對蔗糖合成酶基因家族的調(diào)控作用進(jìn)行了生物信息學(xué)分析。結(jié)果顯示,張雜谷提取物富含多種抗氧化物質(zhì),能夠有效抑制DNA氧化損傷和減輕細(xì)胞內(nèi)氧化應(yīng)激。進(jìn)一步的RNA-seq分析表明,張雜谷提取物顯著調(diào)節(jié)了蔗糖合成酶基因家族的表達(dá),并且通過KEGG通路分析發(fā)現(xiàn),張雜谷提取物可以影響多種碳水化合物代謝通路的活性。此外,我們還利用基因共表達(dá)網(wǎng)絡(luò)構(gòu)建算法,發(fā)現(xiàn)在張雜谷提取物抗氧化過程中,有一些關(guān)鍵基因在不同的代謝途徑中協(xié)同作用,進(jìn)一步驗(yàn)證了張雜谷提取物對代謝和抗氧化的多重作用。本研究為探索張雜谷提取物在膳食中的應(yīng)用提供了新的理論依據(jù)。

Abstract:

ThisstudyinvestigatedtheantioxidativecapacityofZhangzagrainextractanditsregulatoryeffectsonthesucrosesynthasegenefamilythroughbioinformaticsanalysis.TheresultsshowedthatZhangzagrainextractisrichinvariousantioxidativesubstances,effectivelyinhibitsDNAoxidativedamage,andreducesoxidativestressincells.Furthermore,RNA-seqanalysisrevealedthatZhangzagrainextractsignificantlyregulatedtheexpressionofthesucrosesynthasegenefamily,andKEGGpathwayanalysisshowedthatZhangzagrainextractcanaffecttheactivityofmultiplecarbohydratemetabolismpathways.Inaddition,weusedageneco-expressionnetworkconstructionalgorithmandfoundthatsomekeygenesindifferentmetabolicpathwayscooperateduringtheantioxidativeprocessofZhangzagrainextract,furthervalidatingitsmultipleeffectsonmetabolismandantioxidation.ThisstudyprovidesnewtheoreticalevidencefortheapplicationofZhangzagrainextractindiet.

OurresultsshowedthatZhangzagrainextractexhibitedhighantioxidantactivityinvitrobyscavengingfreeradicalsandinhibitinglipidperoxidation.Inaddition,weobservedasignificantincreaseinantioxidantenzymeactivitiessuchascatalase,superoxidedismutase,andglutathioneperoxidaseinthepresenceofZhangzagrainextract,indicatingitspotentialtoenhancetheendogenousantioxidantdefensesystem.

Toexplorethemolecularmechanismsunderlyingtheseeffects,weperformedtranscriptomicanalysisandidentifiedseveralkeygenesinvolvedindifferentmetabolicpathways.Notably,sucrosesynthase,akeyenzymeinvolvedinsucrosemetabolism,wassignificantlyupregulatedinresponsetoZhangzagrainextracttreatment.ThisobservationsuggeststhatZhangzagrainextractmayregulatemetabolicpathways,leadingtothegenerationofenergyandvariousmetabolicproductsthatcontributetotheantioxidativeprocess.

Furthermore,constructionofageneco-expressionnetworkprovidedevidenceofthefunctionalcooperationbetweendifferentmetabolicpathwaysduringtheantioxidativeprocessofZhangzagrainextract.Theidentifiedkeygenefamiliesinthenetworkmayactaspotentialtargetsforfurtherinvestigationandcouldleadtothediscoveryofnewtherapeuticagents.

Insummary,ourstudyhighlightsthepotentialhealthbenefitsofZhangzagrainextractasasourceofnaturalantioxidants.ThefindingsofthisstudyprovideanewperspectivefortheapplicationofZhangzagrainextractasadietarysupplementTheantioxidativepropertiesofZhangzagrainextractcouldhavesignificantimplicationsforhumanhealth.Studieshavesuggestedthatoxidativestressisakeyplayerinthepathogenesisofvariouschronicdiseases,includingcancer,diabetes,andcardiovasculardisorders.Antioxidantshavebeenshowntomitigateoxidativestress,therebypotentiallypreventingordelayingtheonsetofthesediseases.

Inadditiontoitsantioxidativeproperties,Zhangzagrainextractmayalsohaveotherhealth-promotingeffects.Forexample,studieshavedemonstratedthatithasanti-inflammatoryandanti-obesityeffects.Furthermore,Zhangzagrainextracthasbeenshowntomodulategutmicrobiota,potentiallyimprovingguthealthandenhancingimmunefunction.

However,furtherresearchisneededtofullyelucidatethemechanismsunderlyingthehealthbenefitsofZhangzagrainextract.Inaddition,futurestudiesshouldinvestigateitssafetyandefficacyinhumans.Nonetheless,thesefindingssuggestthatZhangzagrainextractcouldbeapromisingsourceofnaturalantioxidantsandpotentiallyotherbioactivecompoundswithpotentialhealthbenefitsInadditiontoitspotentialhealthbenefits,Zhangzagrainextractmayhaveeconomicbenefits.Theextractcouldbeusedinthedevelopmentoffunctionalfoodsanddietarysupplements,offeringamarketableproductthatcouldhelptomeetthegrowingdemandfornaturalhealthproducts.Additionally,thecultivationandprocessingofZhangzagraincouldprovideasourceofincomeforfarmersandagriculturalcommunities,potentiallycontributingtoruraldevelopmentinChinaandotherregionswherethecropisgrown.

However,thedevelopmentandcommercializationofZhangzagrainextractfacesseveralchallenges.Onemajorobstacleisthelackofawarenessandunderstandingofthecropanditspotentialbenefitsamongconsumers,regulators,andthebroaderscientificcommunity.Additionally,theremaybeissuesrelatedtofoodsafety,qualitycontrol,andsourcingofrawmaterialsthatneedtobeaddressed.
