外文原文EffectandMechanismofsomeValidComponentsofChineseHerbsintheLPS-InducedAbortionRatsGraduatestudent:WeiYanmingSupervisor:Prof.ZhaoXingxuPregnancyisacomplexprocessregulatedpreciselyatmultilevelsbynumerousfactorssuchashormones,cytolcinesandothermolecules,however,thefunctionsofmostthesefactorsduringpregnancyhasnotbeenfullyelucidated.Theetiologicalcausesofrecurrentreproductivefailurearediverse,manyofthemarenotevidentinroutineclinicalpracticeandsomearestillunknowntosophisticatedresearchersinthefieldofreproductionmedicine.Generallythecausesofrecurrentpregnancylossareclassifiedasgenetic,endocrinologic,anatomic,immunologic,microbiologic,andenvironmental.Thereismuchevidencethatcytokinesplayavitalroleintheestablishmentandmaintenanceofpregnancybymodulatingimmuneandendocrinesystems.EvidencefrommanystudiesonmarineandhumanpregnancypointstoastrongassociationbetweenmaternalTh2-typeimmunityandsuccessfulpregnancyontheonehandandbetweenThl-typeimmunereactivityandpregnancylossontheother.Th2-typeimmunitycellsmayplayprotectiverolesduringpregnancy.Progesteroneandestrogenarecentraltothemaintenanceofpregnancy,andisthustheidealtargetforfertilityregulation.Progesteroneandestrogenhavebeenknowntoexertextensiveeffectsviaprogesteronereceptor(PR)andestrogenreceptor(ER)ondiversephysiologicalanddevelopmentalfunctionsofthepregnancy,meanwhile,thereceptorexpressionpatternschangedramaticallyduringpregnancyprocess.Chineseherbalmedicineshasbeenusedfortheempiricaltreatmentofvariousdiseases,especiallyrecurrentorhabitualabortion,andreportedtobeclinicallyeffectiveforthesepatientsoranimals.However,themechanismofthetherapeuticeffectsremainsuncertain.Therefore,weinvestigatedtheimmunologicandendocrinologicmechanismofChineseherbsinthetreatmentofabortion.Spragye-Dawley(SD)ratsinjectedwithLipopolysaccharides(LPS)1intailveinonDay7ofpregnancywereusedasabortingmodel.Theratswererandomlydividedintofivegroups:thecontrolgroup,theabortionmodelgroup,thegrouptreatedwiththeAstragalusPolyaccharin,thegrouptreatedwiththeEpimediihe}baandthegrouptreatedwithRadixscutellariaeandRhizomaatractylodisdecoction.Thecontrolgroupandtheabortionmodelgroupweregivennormalsaline2m1fromDay4toDay9ofpregnancyviagastrogavage,allthethreetreatedgroupsweregivenAstragalusPolyaccharin,EpimediiheYbaandRadixscutellariaeandRhizomaatractylodisdecoction2m1respectivelyduringthesameperiod.AlltheratswerekilledonDay10ofpregnancy,abortionratioswereassessedandbloodanduterinetissuesamplesweretakenforthedeterminationofThl/Th2typecytokines.TheconcentrationsofThl/Th2cytokinesinplasmaanduterinetissuesweredeterminedbyenzyme-linkedimmunosorbantassay(ELISA)methodtheERandPRcontentsintheuterinetissueswerealsomeasuredbyELISAmethod.ERmRNAandPRmRNAintheuterinetissuepreparationsweredeterminedbyreversetranscription-polymerisechainreaction(RT-PCR).Theuterinesectionswerepreparedforthehistologicalobservations.Thesignificantfindingsinthecurrentresearchinclude:1.Ahighmiscarriagerateupto72.73%wasobservedintheLPSinducedabortiongroupincontrasttothecontrolgroup,whichwasonly8.33%.2.SignificantlhigherconcentrationsofTh2cytokinesinterleukin-4(IL-4),interleukin-10(IL-10)weremonitoredbothinbloodplasmaanduterinetissuepreparationsoftheratsinthecontrolgroupandthegroupstreatedwiththeChineseherbsthantheratsintheLPSinducedabortionmodelgroup(P<0.01),whilesignificantlyhigherconcentrationsofThlcytokinesinterleukin-2(IL-2),tumornecrosisfactor(TNF)andgamma-interferon(IFN-Y)werefoundintheratsinabortiongroupascomparedtonormalpregnancyratsandtheratstreatedwithChineseherbs(P<0.01),indicatingthatadistinctTh2-biasinnormalpregnancyandaThl-biasinabortionmodel.3.SignificantlyhigherlevelsofERandPRwerefoundintheratsofthenormalgroupandChineseherbstreatedgroupsthantheratsofabortionmodelgroup(P<0.01).4.ThelevelsofERmRNAandPRmRNAexpressioninuterinetissuepreparationsweresignificantlylowerinthemodelgroupthanthecontrolandtheChineseherbstreatedgroups(P<0.01).5.Obviouspathologicalchangesintheuterusescharacterizedbytheemergenceofplentyeosinophilswerefoundinthemodelgroup.6.AbortionrateinducedbyLPSwasdecreasedaftertreatmentwithChineseherbs.AstragalusPolyaccharin,EpimediiherbaandRadixscutellariaeandRhizomaatractylodisdecoction),thesetreatmentcanincreasetheTh2tokinesanddecreasetheThlcytokinesinplasmaanduterinetissuepreparation,thentriggerapredominantTh2responseinthemodel.TheChineseherbsalsocanincreasethecontentsofERandPRintheuterinetissuepreparations,andenhancetheERmRNA,PRmRNAexpressioninuterusesinaboutingrats.TheresultssuggestthatprofileofThl/Th2typecytokinestiltstoTh2immunityinnormalpregnancy,buttoThlimmunityinabortingmodel.Therefore,Chineseherbsseemtobeapotentialmedicineforprotectingpregnancyfromabortion.TheycouldmakeashiftfromThltoTh2immunity,increasetheERandPRcontentsinuteruses,increasetheERmRNAandPRmRNAexpressioninuterusesandreversetheabortioneffectofLPS.Keywords:LPSinducedabortion;Thl/Th2cytokines;ER;PR;AstragalusPolyaccharin;Epimediiherba;RadixscutellariaeandRhizomaatractylodisdecoction;Rats中藥成分對(duì)LPS誘導(dǎo)流產(chǎn)大鼠作用機(jī)理的研究崔雪梅指導(dǎo)老師:翟向和(河北農(nóng)業(yè)大學(xué)中獸醫(yī)學(xué)院定州073000)原作者:魏彥明趙興緒早期胚胎死亡或胚胎丟失,又稱(chēng)隱性流產(chǎn),嚴(yán)重影響母畜繁殖力與生產(chǎn)力。在人醫(yī)臨床上,自然流產(chǎn)的發(fā)生率占全部妊娠15%左右,多數(shù)為早期流產(chǎn)。妊娠時(shí)母體能夠耐受同時(shí)帶有父方遺傳基因與母方遺傳基因的半同種異體胎兒,是極其復(fù)雜而又受到精細(xì)調(diào)控的免疫學(xué)現(xiàn)象。許多證據(jù)表明Thl和Th2細(xì)胞因子在妊娠過(guò)程起著調(diào)節(jié)免疫和內(nèi)分泌的關(guān)鍵作用。雌激素和孕激素在母畜懷孕過(guò)程以至整個(gè)生殖活動(dòng)和其他生理活動(dòng)過(guò)程中均具有重要作用,雌激素受體和孕激素受體則是實(shí)現(xiàn)這些功能所必需的基本元件。為了研究中藥或中藥成分對(duì)實(shí)驗(yàn)性流產(chǎn)大鼠保胎的作用及其機(jī)理,本實(shí)驗(yàn)采用懷孕7天大鼠尾靜脈注射細(xì)菌脂多糖,建立SD大鼠流產(chǎn)模型,再隨機(jī)分為正常對(duì)照組、流產(chǎn)模型組、黃芪多糖組、淫羊藿組和白術(shù)黃苓組。分別于懷孕4~9天口服黃芪多糖、淫羊藿提取物和白術(shù)黃苓2ml/日,對(duì)照組與模型組分別于4~9天口服生理鹽水2m1/日。于實(shí)驗(yàn)第10天麻醉處死大鼠,計(jì)數(shù)妊娠成功率和流產(chǎn)率,并以雙抗體夾心ELISA法檢測(cè)血漿和子宮組織勻漿中Thl型細(xì)胞因子IL-2,TNF-a和IFN-y,Th2型細(xì)胞因子IL-4,IL-10及子宮組織勻漿中ER,PR的含量變化,用RT-PCR法檢測(cè)子宮組織勻漿中ERmRNA和PRmRNA的表達(dá)變化,同時(shí),用光鏡觀察子宮組織結(jié)構(gòu)的病理學(xué)變化,研究黃芪多糖、淫羊藿提取物和白術(shù)黃苓水煎液干預(yù)對(duì)妊娠結(jié)局和以上指標(biāo)的影響。本研究表明:1.LPS誘導(dǎo)大鼠流產(chǎn)模型流產(chǎn)率高達(dá)72.73%,對(duì)照組為8.33%,兩者差異極顯著(P<0.01)。流產(chǎn)模型組大鼠血漿和子宮組織勻漿Th1型細(xì)胞因子IL-2、TNF-a和IFN-y含量顯著增加,Th2型細(xì)胞因子IL-4、IL-10含量則顯著降低,與對(duì)照組比較差異極顯著(P<0.01)。流產(chǎn)模型組子宮組織中ER,PR
