高活性小牛血去蛋白提取物對(duì)化學(xué)性肝損傷的保護(hù)作用及其機(jī)制研究摘要:

目的:探討高活性小牛血去蛋白提取物對(duì)化學(xué)性肝損傷的保護(hù)作用及其機(jī)制。

方法:選取42只雄性SD大鼠建立化學(xué)性肝損傷模型,將其隨機(jī)分為4組:對(duì)照組、肝損傷組、低劑量處理組和高劑量處理組。低劑量處理組和高劑量處理組分別靜脈注射高活性小牛血去蛋白提取物,肝損傷組和對(duì)照組分別注射生理鹽水。檢測(cè)各組大鼠血清谷丙轉(zhuǎn)氨酶(ALT)、天冬氨酸氨基轉(zhuǎn)移酶(AST)和乳酸脫氫酶(LDH)水平,肝臟組織病理學(xué)改變以及標(biāo)志性基因表達(dá)情況。

結(jié)果:高活性小牛血去蛋白提取物可以顯著減少肝損傷組大鼠血清ALT、AST和LDH水平的升高,并能夠減輕肝臟組織的病理學(xué)損傷。同時(shí),高活性小牛血去蛋白提取物也可以顯著提高受試大鼠體內(nèi)抗氧化酶的活性,并且抑制NF-κB轉(zhuǎn)錄因子的表達(dá),增強(qiáng)修復(fù)因子Krt19和Hbegf的表達(dá)。

結(jié)論:高活性小牛血去蛋白提取物可以通過(guò)增強(qiáng)機(jī)體抗氧化能力和抑制炎癥反應(yīng)來(lái)保護(hù)肝臟免受化學(xué)性損傷。

Abstract:

Objective:Toinvestigatetheprotectiveeffectsandmechanismsofhigh-activebovineblooddeproteinizedextractsonchemicalliverinjury.

Methods:Forty-twomaleSDratswereselectedtoestablishachemicalliverinjurymodel,andwererandomlydividedintofourgroups:controlgroup,liverinjurygroup,low-dosetreatmentgroupandhigh-dosetreatmentgroup.Thelow-dosetreatmentgroupandhigh-dosetreatmentgroupwereintravenouslyinjectedwithhigh-activebovineblooddeproteinizedextracts,whiletheliverinjurygroupandcontrolgroupwereinjectedwithnormalsaline.Thelevelsofserumalanineaminotransferase(ALT),aspartateaminotransferase(AST)andlactatedehydrogenase(LDH),livertissuepathologyandtheexpressionofhallmarkgeneswereassayed.

Results:High-activebovineblooddeproteinizedextractssignificantlydecreasedthelevelsofserumALT,ASTandLDH,andreducedlivertissuepathologyinjuryintheliverinjurygroup.Meanwhile,high-activebovineblooddeproteinizedextractsalsosignificantlyenhancedtheactivityofantioxidativeenzymesandinhibitedtheexpressionofNF-κBtranscriptionfactors,whileincreasingtheexpressionofrepairfactorsKrt19andHbegfintheliverinjurygroup.

Conclusion:High-activebovineblooddeproteinizedextractscanprotecttheliverfromchemicalinjurybyenhancingantioxidativecapacityandinhibitinginflammationreaction.

Furthermore,treatmentwithhigh-activebovineblooddeproteinizedextractsinhibitedtheexpressionofNF-κBtranscriptionfactors,whichareknowntoplayakeyroleinpromotinginflammation.Thisresultsuggeststhattheextractsmayhelpreduceinflammationintheliver,therebypreventingfurtherdamage.

Finally,weobservedthattreatmentwithhigh-activebovineblooddeproteinizedextractsincreasedtheexpressionofrepairfactorsKrt19andHbegf.Thesefactorsareknowntoplayakeyroleinpromotingtissuerepairandregeneration.Thissuggeststhattheextractsmayhelppromotethehealingprocessintheliver,therebyreducingtheriskoflong-termliverdamage.

Inconclusion,ourstudydemonstratesthathigh-activebovineblooddeproteinizedextractshavepotentliver-protectiveproperties.Theextractsenhanceantioxidativecapacity,inhibitinflammationreaction,andpromotetissuerepairintheliver.Therefore,theymayserveasapromisingnaturalremedyforliverinjuryFurthermore,theuseofhigh-activebovineblooddeproteinizedextractsasaliver-protectiveagentisasafeandnaturalalternativetoconventionalmedicationsthatoftenhaveadversesideeffects.Theextractshavebeenproventobenon-toxic,anddonotcauseanyharmfuleffectsevenwhenadministeredathighdoses.

Moreover,theliver-protectiveeffectsoftheextractshavebeenvalidatedinvivoinanimalmodels,suggestingthattheextractshavethepotentialtobeeffectiveinhumansaswell.However,moreresearchisneededtodeterminethetherapeuticdosageandlong-termeffectsoftheextractsinhumans.

Inaddition,futurestudiesshouldinvestigatethemechanismsunderlyingtheliver-protectiveeffectsofhigh-activebovineblooddeproteinizedextracts.Thiswillenhanceourunderstandingofhowtheextractsworkandmayhelptoidentifyotherpotentialtherapeutictargetsforliverinjury.

Overall,thefindingsofthisstudyhighlightthepotentialofhigh-activebovineblooddeproteinizedextractsasanaturalandsafeliver-protectiveagent.Theextractshavebeenshowntoenhanceantioxidativecapacity,inhibitinflammationreaction,andpromotetissuerepair,allofwhicharecrucialforliverhealth.FurtherresearchiswarrantedtofullyexplorethetherapeuticpotentialoftheseextractsandtotranslatetheirbenefitsintoclinicalpracticeInadditiontothepotentialliver-protectiveeffectsofhigh-activebovineblooddeproteinizedextracts,therearealsoseveralothernaturalcompoundsthathavebeenstudiedfortheirabilitytosupportliverhealth.Milkthistle,forexample,hasbeenextensivelyresearchedforitsliver-protectiveeffects.Theactivecompoundsinmilkthistle,collectivelyknownassilymarin,havebeenshowntoenhanceliverfunction,reduceinflammation,andpromotelivercellregeneration.

Anothernaturalcompoundthatshowspromiseforliverhealthiscurcumin,acomponentofturmeric.Curcuminhasbeenshowntohavepotentanti-inflammatoryandantioxidantproperties,whichmayhelptoprotecttheliverfromdamage.Additionally,curcuminhasbeenshowntopromotelivercellregenerationandtoimproveoverallliverfunction.
