目錄摘要 MMRN1對PTMC遷移和增殖的研究摘要目的:探討MMRN1對PTMC(甲狀腺微小乳頭狀癌,屬于甲狀腺乳頭狀癌的一類,癌直徑≤10?mm)轉(zhuǎn)移和增殖作用的研究。方法:查詢PTMC轉(zhuǎn)移和增殖的差異性序列并篩選差異性基因;通過免疫組織化學(xué),WesternBlot,qPCR等方法測定PTC細胞和正常細胞中MMRN1的表達情況;使用細胞劃痕實驗驗證MMRN1對PTC細胞遷徙的影響;利用細胞成團實驗判斷MMRN1對PTC細胞增殖的影響。結(jié)果:篩選到PTMC中顯著表達差異基因MMRN1;免疫組織化學(xué),WesternBlot,qPCR等結(jié)果均顯示MMRN1在PTC細胞中高表達,且在BCPAP細胞中高表達于TPC-1細胞;WesternBlot結(jié)果顯示相比于正常甲狀腺細胞,BCPAP細胞中MMRN1的表達含量高64%,且相比于TPC-1細胞,BCPAP細胞中MMRN1的表達高39%;qPCR結(jié)果顯示相比于正常甲狀腺細胞,BCPAP細胞中MMRN1的表達含量高88%,且相比于TPC-1細胞,BCPAP細胞中MMRN1的表達高48%;細胞劃痕實驗結(jié)果顯示,PTC細胞遷徙能力強于正常甲狀腺細胞,且BCPAP細胞的遷徙能力強于TPC-1;細胞成團結(jié)果顯示,BCPAP成團聚集能力強于TPC-1,TPC-1成團能力強于正常甲狀腺細胞。結(jié)論:MMRN1與PTMC發(fā)生有關(guān),并可能促使其遷移與增殖。關(guān)鍵詞MMRN1;甲狀腺癌細胞;增殖;遷移;
