TheoperatingprincipleoftheHEMOXANALYZERisbasedondualwavelengthspectrophotometryforthemeasurementoftheopticalpropertiesofhemoglobinandanoxygenelectrodeformeasuringtheoxygenpartialpressureinmillimetersofmercury.TheresultingsignalsfrombothmeasuringsystemsarefedtotheXYrecorderorPC,whichplotstheresultingcurve.

Afterthesamplebuffersolutionhasbeendrawnintothecuvette,thetemperatureofthemixturehastoequilibrateuntilithasreached37°C.Thanthesampleisoxygenatedwithairoroxygen,suppliedfromanoutsideairtank.AfterthesamplehasbeencompletelyoxygenatedandthepO2hasbeenadjustedtothecorrectvalue,theplotingofthedeoxygenationcurvecanbeginbyswitchingtheAIR/NITROGRENswitchtoNITROGEN.ThecompletionofthecurveisreachedwhenthepO2approacheszero.

Thetimeforcompletingtheplottingofthecurveisapproximately15-20minutesbutmaybereducedbyincreasingtheflowofnitrogen.

Themeasurementisnormallycarriedoutatatemperatureof37°Ctosimulatephysiologicalconditions.However,thetemperaturecontrolsystempermitsmeasurementatothertemperaturesinconjunctionwiththebuiltincoolingcoilconnectedtoanexternalHeating/Coolingbath(suppliedbythecustomer).

TheHEMOXAnalyzerconsistsofthemainconsolewhichisasheetmetalhousing,uponthebaseofwhichtheopticalandelectroniccomponentsaremounted.Alsocontainedinthehousingisthegasselectionvalvesystemandthepumpingsystemforsampleintakeandremoval.

Thehousingcontainsinitsfrontsectiontheopticalbenchwithlightsource,lenssystem,cuvetteholderandthephotomultiplierdetectors.Therearpartofthehousingcontainsthepowersuppliesandtheelectronicchassis,includingthefourPCboards.ThehousingcoverhasasmallhingedlidforeasyaccesstothesamplecuvetteforsampleloadingandroutinemaintenanceofthecavityandthepO2electrode.

Thecompletetopcoverofthehousingishingedontherightsideofthebaseandcanbeswungopenbyremovaloftwonuts,locatedunderthebaseofthehousing.Allutilities,likemainpower,gasesandoptionalcoolingwater,enterfromtherearoftheinstrument.Alsothemainfuse(2.0amps.)islocatedintherear.Alloperationalcontrolsarefrontpanelmountedforeasyaccessandconvenienceofadjustment.

TheDAQcomesHEMOXanalyzerandisuniquelycalibratedtoit.TheHASreadsthedatapointscapturedviatheDAQandplotstheOxygenEquilibriumCurveinbothoxyanddeoxystate.Thep50,PO2,S1/S2(logPO2),andTemperaturearedisplayedinreal-time.Whenthetestrunhascompletedtheestimatedp50willdoaregressionanalysistocalculatetheactualp50.TheuserhastheoptionofviewingnumerousotherresultssuchastheHillPlotandHillDerivativeCurve.ConvergenceofAdairconstantsandadatatablewithp0throughp100isalsoviewable.Theusercansetthecooperativityrangewithapreferencesectionandcanalsochoosehowtosmooththecurvefittingprocess.Theycanalsochooselinearorlogarithmicmodesalongwithoverallorstepwiseprocessing.
